I need to run my outdoor landscape lights 300 to 500 feet. Can I do that using only one transformer?

Yes you can use one transformer that is sized to the total wattage of the load. Your problem will be voltage drop due to the distance you have described. I would not recommend that you go past 200 feet per run without the use of a multi-tap transformer that has a 13 volt output and #10 or #8 wire. Here is a link to one of those products:
